Abstract:
This invention discloses a method for preparing polyclonal antibody against Escherichia coli and its application in detecting Escherichia coli. The method comprises: (1) separating Escherichia coli from domestic wastewater; (2) culturing and inactivating to obtain whole somatic antigen of Escherichia coli; (3) immunizing animals with the antigen; (4) separating and purifying antiserum from the immunized animals to obtain the polyclonal antibody against Escherichia coli. The obtained polyclonal antibody has such advantages as high specificity, high purity, high titer (higher than 1: 1*105) and long storage time, and can be used in the immuno-detection of Escherichia coli in the environment or food. The immuno-detection method has such advantages as high accuracy, high sensitivity and few detection procedures (by using unbroken bacterium as the antigen).
