Abstract: |
The present invention discloses one method of raising methanol tolerance of saccharomycete, and the method is to introduce polyhydroxy fatty acid ester synthesizing way related gene into saccharomycete. The present invention introduce polyhydroxy fatty acid ester synthesizing way related gene into Pichia pastoris GS115 to obtain saccharomycete phenotypic mutant strain with fast growth and raised methanol tolerance. By means of combined gene engineering and microbial fermentation method, the present invention optimizes the physiological metabolism wary of saccharomycete creatively to promote the growth of saccharomycete and provide excellent host strain for low cost and high protein output. Further, the present invention illuminates the important significance of the creation of PHA on the metabolism and the physiological function of host and the benefit of foreign introduction of PHA biosynthesizing way on host. |
