Abstract:
The invention relates to a fluorescence correlation spectrometer of gradient field. The spectrometer is composed of object lens of microscope, focusing lens, laser attenuation piece, detector for single atom, signal collecting system and signal processing system etc., also second two-color beam splitter and infrared laser attenuation piece. Fluorescence exciting light via exciting laser attenuation piece. then is reflected by second two-color beam splitter, at last the light beam is focused to a point on a specimen measured. It will excite the molecules in said point to produce fluorescence. This fluorescence is collected by object lens, then it is reflected, filtered, focused to confocal pinhole. The going beam is received by single photon control-detector to produce photon-receiving signal entering a processing system. After passing through attenuation piece and second two-color beam splitter, infrared laser beam joints to exciting laser beam. Passing through first two-color beam splitter the jointed beam is focused to the above said same point in the specimen. Thus produced laser gradient field produces controlling action on molecular movement of this point in the specimen. The invented instrument can accomplish all functions of present fluorescence correlation spectrometer, and can measure polarizability of particle and its variety.
