Abstract: |
The present invention discloses a identification process of the alleles. In this process, with the DNA from the sample as the template, the primer group for the corresponding alleles of one or more mutations of the target genes and the DNA polymerase without 5'-3'exonuclease activity, the allelic fragments of the target genes containing the mutant site are amplified via mutiplex PCR. The PCR products are hybridized with general chip, the allele is determined by the result of hybridization. The primer group includes a general primer, two allele-specific primers and a common primer corresponding to each mutant site of the target gene. All the allel-specific primers are each connected to a different Tag sequence at the 5' end. The above general chip contains several Tag probes, each is the same with the corresponding Tag sequence. Every Tag probe is hybridized with the complement sequence in the multiplex PCR products. |
